For use as first aid to help prevent infection in minor cuts, scrapes, and burns.
Clean the affected area and apply a small amount of the product 1 to 3 times daily. The area may be covered with a sterile bandage; if bandaged, allow the product to dry first.
Dilute each dose and administer intravenously by infusion over 60 minutes, using sodium chloride or glucose solution as the diluent. Do not mix cloxacillin with other drugs in the same infusion. Switch to oral therapy as soon as clinically appropriate.
Cloxacillin is a penicillinase-resistant penicillin belonging to the beta-lactam class of antibiotics. It works by inhibiting bacterial cell wall synthesis. Resistance to penicillinase (beta-lactamase) is conferred by a bulky substituent on the beta-lactam ring, which prevents the enzyme from binding to and inactivating the drug — making cloxacillin effective against penicillinase-producing staphylococci that are resistant to standard penicillins.
Following parenteral administration, cloxacillin distributes throughout body tissues and fluids, achieving therapeutic concentrations in skin, lungs, bones, synovial fluid, urine, and the gallbladder. The serum half-life is approximately 30 minutes. The drug is primarily eliminated by the kidneys.
Gastrointestinal disturbances (particularly diarrhoea) and allergic reactions — which may occasionally be severe — have been reported. Haematological disorders occur rarely.
Do not use in the eyes or over large areas of the body. Do not use if you are allergic to povidone-iodine or any other ingredient in this preparation.
For external use only.
Experience in this population is limited. Cautious administration and frequent monitoring of organ system function are recommended.
Cloxacillin is primarily eliminated by the kidneys. Owing to its low inherent toxicity, dose adjustment is generally not required; however, a dose reduction should be considered in patients with severely impaired renal function.
Cloxacillin has been assigned to pregnancy category B. No controlled data in human pregnancies are available, and no reports of congenital abnormalities associated with its use have been identified in the literature. It should be used during pregnancy only when clearly necessary.
Cloxacillin produces low levels in breast milk that are not expected to cause adverse effects in breastfed infants. Following a 500 mg intramuscular dose, milk concentrations ranged from 0.1 to 0.3 mg/L at 1 to 6 hours after the dose. Occasional diarrhoea has been reported in breastfed infants of mothers receiving cloxacillin; no rashes or candidiasis were observed. Cloxacillin is considered acceptable for use in nursing mothers.
In the event of a suspected overdose, seek emergency medical care or contact a poison control centre immediately.
Do not combine with methotrexate, as cloxacillin may increase methotrexate toxicity.
Store below 25 °C in a dry place. Once reconstituted, the solution must be used immediately.
