Vitamin C plays an important role in the body's response to stress and in defence against infection. It possesses some anti-inflammatory activity, protects cells against oxidative damage, and enhances iron absorption from the gastrointestinal tract.
Vitamin E's primary role is the prevention of oxidation of polyunsaturated fatty acids. It reacts with free radicals — which cause oxidative damage to cell membranes — without generating a new free radical in the process.

Beta carotene in this tablet is converted to Vitamin A (Retinol) as the body requires it. Retinol serves several biochemical functions — in the retina, in growth, in tissue differentiation, and in immunological response. It also has some anti-cancer activity.
Vitamin C is the most powerful reducing agent known to be present in living tissues; its deficiency causes scurvy. It acts as a cofactor in numerous biological processes. Vitamin C and molecular oxygen are essential for the conversion of proline to hydroxyproline and of dopamine to noradrenaline. Vitamin C is also essential for the synthesis of adrenal steroid hormones. It plays an important role in defence against infection, supporting the normal functioning of T-lymphocytes and leukocytes. Ascorbic acid has some anti-inflammatory activity and protects cells against oxidation of essential molecules. At high doses (1–2 g daily), ascorbic acid increases iron absorption.
Vitamin E acts as a defence against oxidative stress and lipid peroxidation. In most cell membranes there is approximately one molecule of tocopherol for every 1,000 lipid molecules. Tocopherol neutralises peroxide radicals and then requires a supply of reduced hydrogen — usually provided by ascorbic acid or reduced glutathione — to restore the steady-state condition.
Loose stools may occasionally occur, and the skin may develop a slight yellow discolouration. Any toxic effects of Vitamin A (produced by conversion from beta carotene) are normally reversible; irreversible toxicity is effectively unknown. Chronic overdosage may lead to peeling and redness of the skin, disturbed hair growth, loss of appetite, and nausea.
Vitamin C is generally well tolerated. Large doses have been reported to cause diarrhoea and other gastrointestinal disturbances. High doses may also lead to hyperoxaluria and the formation of renal calcium oxalate stones; it should therefore be used with care in patients with hyperoxaluria.
Vitamin E is generally well tolerated. Large doses may cause diarrhoea, abdominal pain, and other gastrointestinal disturbances, and have also been reported to cause fatigue and weakness.
Although the body converts beta carotene to Vitamin A only as needed, excessive Vitamin A intake should be avoided during pregnancy due to potential teratogenic effects. Patients with hypersensitivity to retinol should not take this preparation, though such cases are rare.
Megadoses of Vitamin C are contraindicated in patients with hyperoxaluria. Pregnant women taking more than 5 g of ascorbate daily have been reported to suffer fetal abortion, and higher doses have been reported to cause failure of conception.
There are no absolute contraindications to Vitamin E.
There is evidence that beta carotene may be harmful to heavy smokers and people who consume alcohol heavily. Caution should be exercised in these groups.
Vitamin C should be given with caution to patients with hyperoxaluria, due to the risk of renal calcium oxalate stone formation.

The toxicity of beta carotene is low. Chronic high intake may cause hypercarotenemia (yellowing of the skin). Because conversion of beta carotene to retinol is not rapid, acute Vitamin A toxicity from beta carotene is unlikely. Prolonged overdosage may lead to peeling and redness of the skin, disturbed hair growth, loss of appetite, and nausea.
Chronic toxic effects can occur with daily intake exceeding approximately 2 g. Symptoms may include weakness, dizziness, diarrhoea, nausea, vomiting, headache, insomnia, heartburn, and skin reactions. Prolonged high-dose use has also been associated with kidney disease, gastritis, stomach ulcers, and disturbances of the pancreas. High doses may cause hyperoxaluria and renal calcium oxalate stone formation.
The toxicity of Vitamin E is low and no mutagenic, teratogenic, or carcinogenic effects are established. Large doses may cause diarrhoea, abdominal pain, fatigue, and weakness. High doses are inadvisable in patients with Vitamin K-associated blood coagulation disorders, as Vitamin E may enhance anticoagulant activity.
No potentially hazardous drug interactions are known for retinol after conversion from beta carotene. Cadmium and copper both decrease retinol plasma levels. Among antibiotics, neomycin and bleomycin reduce retinol absorption.
Vitamin C is incompatible in solution with aminophylline, bleomycin, erythromycin lactobionate, nafcillin, nitrofurantoin sodium, conjugated oestrogens, sodium bicarbonate, sulphafurazole diethanolamine, chloramphenicol sodium succinate, chlorothiazide sodium, and hydrocortisone sodium succinate. It increases the apparent half-life of paracetamol and enhances iron absorption from the gastrointestinal tract.
