A-B1 is indicated for the treatment of various manifestations of thiamine deficiency, including Beriberi, Wernicke's encephalopathy, neuritis associated with pregnancy, and pellagra.

Prophylactic supplementation may also be appropriate in conditions where dietary thiamine intake is low, gastrointestinal absorption is impaired (e.g. due to alcohol use), or physiological requirements are increased (e.g. during pregnancy or with a carbohydrate-rich diet).

Doses of up to 30 mg daily, taken as a single dose or in divided doses, are recommended.

Thiamine Hydrochloride (Vitamin B1) is essential for the metabolism of carbohydrates and many amino acids. In thiamine deficiency, the utilisation of pyruvic acid and certain amino acids in body tissues is reduced, leading to accumulation of pyruvic acid and lactic acid — the underlying mechanism of Beriberi.

In adults, approximately 1 mg of thiamine is fully metabolised by the tissues each day, representing roughly the minimum daily requirement.

Thiamine is well absorbed from the gastrointestinal tract and is widely distributed throughout body tissues. Any amount in excess of the body's requirements is excreted in the urine, either unchanged or as metabolites.

No adverse effects are expected when A-B1 is taken orally, even at doses many times greater than the therapeutic amount.

Hypersensitivity to thiamine.

Serious allergic reactions to thiamine are very rare. Seek immediate medical attention if any of the following symptoms of a severe allergic reaction occur:

• Rash or itching
• Swelling, particularly of the face, tongue, or throat
• Severe dizziness
• Difficulty breathing

A-B1 may be used safely across all population groups, including neonates, children, pregnant women, breastfeeding women, and elderly patients, without additional restriction.

Overdose with Thiamine Hydrochloride does not cause toxicity.
